I have found posts about similar situations, but not any that apply to my
specific situation. I have a 2003 forest that has been preped for 2003 32-bit
R2. Currently there is one 32-bit DC running R2 in the forest. Several 2003
64-bit R2 servers will be promoted to DC's in the forest soon. Since the
forest has been prepped for 32-bit R2 do I need to prep it again before
promoting the 64-bit R2 servers?

As Marcin implied, operating system bit levels (x86 or x64) have nothing to
do with Schema versions, only operating system versions.

There is no difference in the schema between the OS architectures, so No
is the answer to your question.

When you prep the forest you are adding additional schema attributes for the
AD database. This is a multimaster database that both 32 bit and 64 bit
o/s's read and write to. The o/s itself houses the database but AD only
needs to extend the schema once and all changes are replicated to all Domain
Controllers, whether they are 32 bit or 64 bit. So there is no need to run
this a second time but if you do it will just ignore it and tell you already
done.
